Norovirus outbreaks are associated with the consumption of contaminated shellfish, and so efficient methods to recover and detect infectious norovirus in shellfish are important. The Proteinase K digestion method used to recover norovirus from shellfish, as described in the ISO 15216, would be a good candidate but its impact on the virus capsid integrity and thus infectivity was never examined. The aim of this study was to assess the impact of the Proteinase K digestion method, and of the heat treatment component of the method alone, on norovirus (genogroups I and II) and MS2 bacteriophage capsid integrity. A slightly modified version of the ISO method was used. RT-qPCR was used for virus detection following digestion of accessible viral RNA using RNases. MS2 phage infectivity was measured using a plaque assay. The effect of shellfish digestive glands (DG) on recovery was evaluated. In the presence of shellfish DG, a reduction in MS2 phage infectivity of about 1 log10 was observed after the Proteinase K digestion method and after heat treatment component alone. For norovirus GII and MS2 phage, there was no significant loss of genome following the Proteinase K digestion method but there was a significant 0.24 log10 loss of norovirus GI. In the absence of shellfish DG, the reduction in MS2 phage infectivity was about 2 log10, with the addition of RNases resulting in a significant loss of genome for all tested viruses following complete Proteinase K digestion method and the heat treatment alone. While some protective effect from the shellfish DG on viruses was observed, the impact on capsid integrity and infectivity suggests that this method, while suitable for norovirus genome detection, may not completely preserve virus infectivity.  相似文献

We identified different distributions of marine nonindigenous species (NIS) and native species on some artificial structures versus natural reefs and using experimental manipulations, revealed some possible causal mechanisms. In well-established subtidal assemblages, numbers of NIS were 1.5–2.5 times greater on pontoons or pilings than on rocky reefs, despite the local species pool of natives being up to 2.5 times greater than that of NIS. Conversely, on reefs and seawalls, numbers of native species were up to three times greater than numbers of NIS. Differential recruitment to different positions and types of surfaces appeared to influence distribution patterns. NIS recruited well to most surfaces, particularly concrete surfaces near the surface of the water, whilst natives occurred infrequently on wooden surfaces. The position of rocky reefs and seawalls close to the shore and to the seabed appeared to make them favourable for the recruitment of natives, but this positioning alone does not hinder the recruitment of NIS. We argue that pontoons and pilings represent beachheads (i.e. entry points for invasion) for many nonindigenous epibiota and so enhance the spread and establishment of NIS in estuaries. Habitat creation in estuaries may, therefore, be a serious threat to native biodiversity. Spatial autocorrelation correlograms based on Moran's coefficient were constructed for common polychaetes (Goniada emerita, Heteromastus filiformis, Macroclymenella stewartensis, Boccardia spp. and Magelona? dakini)_and bivalves (Nucula hartvigiana, Soletellina siliqua and Tellina liliana) collected from intertidal sandflats of Manukau Harbour (New Zealand) during October, 1987. Patterns of heterogeneity on a scale smaller than inter-sample distance, homogenous density patches (5 to 30 m radius) and gradients in abundance running through sample sites (9 000 m2) were identified. Patterns could be defined even for species with distributions which, based on the variance: mean ratio test, were not significantly different from random. The possible influence on two of the study sites of sediment disturbances generated by feeding rays is discussed. Identification of spatial patterns is considered an important aspect of the design of surveys and manipulative field experiments.  相似文献
